The immune system is meant to protect the body, but for more than 15 million Americans, it turns against them instead.

Today’s therapies treat the damage rather than its source. Most dampen the immune system broadly or block the inflammatory chemicals it releases, while the rogue immune cells producing those chemicals stay in the body. When treatment stops, symptoms return. A newer wave of therapies reprograms immune cells inside the body, but most target B cells through an indirect, multi-step process, while many autoimmune diseases are driven largely by T cells.

ai3Bio’s name nods to the three disciplines it brings to that problem: advanced immunology, autoimmunity, and artificial intelligence. Using its AI platform to analyze diseased tissue, the team found a marker shared by disease-causing Th17 cells across many conditions, giving it a precise way to target them in diseases that have long been treated as unrelated.

It is advancing two complementary platforms to do this. STARx uses a lipid nanoparticle, a tiny fat-based capsule, to deliver mRNA instructions that prompt Th17 cells to self-destruct without triggering more inflammation. T Deplete uses a targeted antibody that flags the same cells for the body’s own immune defenses to clear.

Early results are encouraging. In lab tests on blood samples from autoimmune patients, STARx removed disease-causing Th17 cells. T Deplete showed potent efficacy in non-human primates and is now in the safety studies required before human trials.

The company is led by CEO and repeat biotech entrepreneur Dr. Steven Altschuler; scientific co-founder Dr. Jonathan Kagan of Harvard Medical School and Boston Children’s Hospital; and Chief Technology Officer Dr. Michael Lyman, a drug-discovery veteran who has brought therapeutic antibodies to the clinic. Its scientific advisory board pairs Alnylam founding CEO John Maraganore with leading immunologists, including several members of the National Academy of Sciences. ai3Bio launched with $48 million in Series A funding led by UPMC Enterprises and Ziff Capital Partners.
